چکیده

Protoplasts, RNA, Plant Viruses, Tobacco Mosaic Virus, Polyethylene Glycol Infection of tobacco protoplasts with R N A of tobacco mosaic virus and the bromoviruses has been studied. The quality of the R N A was very important. Ribonuclease activity was controlled by incorporating 5 mM ZnCl2 in the inoculum. Up to 60% infection has been observed with bromovirus R N A but infection with TM V R N A was much less efficient. A procedure for inoculating in 40% polyethylene glycol was developed. High levels of infection were obtained with all the viruses and viral R N A examined.

Infection of Protoplasts from Several Brassica Species with Cauliflower Mosaic Virus Following Inoculation Using Polyethylene Glycol

Using a polyethylene glycol-mediated uptake procedure, protoplasts from a range of Cruciferous species were infected with cauliflower mosaic virus; they include Moricandia arvensis, a plant not k n o w n to be susceptible. With this technique > 90~o of turnip protoplasts were infected, each producing an average of 104 to 105 viral genome equivalents. Filter hybridization and fluorescent antibod...

A simplified Protocol to Induce Callogenesis in Protoplasts of Date Palm (Phoenix dactylifera L.) Cultivars

Background: In Algeria, date palm is currently confronted to the Bayoud disease. Biotechnological tools such as protoplasts fusion can appear as an alternative to ensure rapid multiplication and improvement of this species. Objectives: Callogenesis induction in protoplasts isolated from embryogenic callus of three date palm cultivars. Materials and Methods: Some factors influencing the isolatio...

The plant defense response to cucumber mosaic virus in cowpea is elicited by the viral polymerase gene and affects virus accumulation in single cells.

Resistance to infection in cowpea by strains of cucumber mosaic virus (CMV) involves a local, hypersensitive response (HR) and a localization of infection. These responses can be separated by mutation at two sites (nucleotides 1978 and 2007, in codons 631 and 641) in the CMV 2a polymerase gene. Changes to both sites of a restricted strain allow systemic infection without an HR and increase the ...

PLANT REGENERATION FROM MESOPHYLL PROTOPLASTS OF POTATO (SOLANUM TUBEROSUM L.) CULTIVAR DELAWARE USING SILVER THIOSULFATE (STS)

The use of in vitro potato shoot cultures for protoplast isolation is desirable for consistent protoplast qualities. Ethylene build-up in the culture vessels causes problems for leaf growth (e.g. decreasing leaf surface, dry weight and chlorophyll) in shoot culture. Fifty to one hundred µM Silver thiosulfate (STS) produced a larger leaf area as a source of plant material for protoplast isolatio...

Salicylic acid has cell-specific effects on tobacco mosaic virus replication and cell-to-cell movement.

Tobacco mosaic virus (TMV) and Cucumber mosaic virus expressing green fluorescent protein (GFP) were used to probe the effects of salicylic acid (SA) on the cell biology of viral infection. Treatment of tobacco with SA restricted TMV.GFP to single-epidermal cell infection sites for at least 6 d post inoculation but did not affect infection sites of Cucumber mosaic virus expressing GFP. Microinj...
